This variant lies in the ALMS1 gene (transcript NM_001378454.1) at coding-DNA position 2471, where C is replaced by A; at the protein level this means replaces alanine at residue 824 with aspartic acid — a missense variant. Submitter rationale: Variant summary: ALMS1 c.2468C>A/p.Ala823Asp (also known as c.2474C>A/p.Ala825Asp in RefSeq) results in a non-conservative amino acid change in the encoded protein sequence. Three of four in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 8e-06 in 248980 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance.
