Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_024675.4(PALB2):c.1424C>T (p.Ser475Leu), citing Ambry Variant Classification Scheme 2023: The p.S475L variant (also known as c.1424C>T), located in coding exon 4 of the PALB2 gene, results from a C to T substitution at nucleotide position 1424. The serine at codon 475 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
