Pathogenic for Aortic aneurysm, familial thoracic 6 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001613.4(ACTA2):c.350A>G (p.Asn117Ser), citing Invitae Variant Classification Sherloc (09022015): ClinVar contains an entry for this variant (Variation ID: 1329099). This sequence change replaces asparagine, which is neutral and polar, with serine, which is neutral and polar, at codon 117 of the ACTA2 protein (p.Asn117Ser). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individuals with thoracic aortic aneurysm and/or dissection or smooth muscle dysfunction (PMID: 27146836, 29202781, 32093627; Invitae). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt ACTA2 protein function. For these reasons, this variant has been classified as Pathogenic.
