Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000810.4(GABRA5):c.86+1G>A, citing LabCorp Variant Classification Summary - May 2015: Variant summary: GABRA5 c.86+1G>A is located in a canonical splice-site and is predicted to affect mRNA splicing resulting in a significantly altered protein due to either exon skipping, shortening, or inclusion of intronic material. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing, however current evidence is not sufficient to establish loss of function as a mechanism for disease. Computational tools predict a significant impact on normal splicing: Three predict the variant abolishes a canonical 5' splicing donor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 4e-06 in 249224 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.86+1G>A has been reported in the literature in an individual affected with familial focal epilepsy (Oates_2018). This report does not provide unequivocal conclusions about association of the variant with Developmental And Epileptic Encephalopathy, 79. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
