Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001386140.1(MTTP):c.1655C>T (p.Pro552Leu), citing LabCorp Variant Classification Summary - May 2015: Variant summary: MTTP c.1655C>T (p.Pro552Leu) results in a non-conservative amino acid change located in the Lipid transport protein, N-terminal domain (IPR001747) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 251132 control chromosomes. c.1655C>T has been reported in the literature as a homozygous genotype in at-least one individual affected with Abetalipoproteinaemia (Bassen-Kornzweig Syndrome) with subsequent cohort overlap (example, Aminoff_2012, Di-Filippo_2014). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as VUS-possibly pathogenic.
