Submitter rationale: Variant summary: NPC1 c.2054T>C (p.Ile685Thr) results in a non-conservative amino acid change located in the Sterol-sensing domain (IPR000731) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251456 control chromosomes. c.2054T>C has been reported in trans with two different pathogenic variants in the literature in individuals affected with Niemann-Pick Disease Type C (example, Wang_2020, Yang_2015, Zhang_2014). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
