Uncertain significance for Axenfeld-Rieger syndrome type 3 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001453.3(FOXC1):c.1337_1342dup (p.Gly447_Gly448insAspGly), citing Invitae Variant Classification Sherloc (09022015): This variant, c.1337_1342dup, results in the insertion of 2 amino acid(s) of the FOXC1 protein (p.Gly447_Gly448insAspGly), but otherwise preserves the integrity of the reading frame. This variant is present in population databases (rs756037576, gnomAD 0.04%), and has an allele count higher than expected for a pathogenic variant. This variant has not been reported in the literature in individuals affected with FOXC1-related conditions. ClinVar contains an entry for this variant (Variation ID: 132835). Algorithms developed to predict the effect of variants on gene product structure and function are not available or were not evaluated for this variant. Experimental studies have shown that this variant does not substantially affect FOXC1 function (PMID: 25786029). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
