This variant lies in the CAPN3 gene (transcript NM_000070.3) at coding-DNA position 966, where T is replaced by A; at the protein level this means converts the codon for tyrosine at residue 322 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: Variant summary: CAPN3 c.966T>A (p.Tyr322X) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, which are commonly known mechanisms for disease. The variant was absent in 251248 control chromosomes. c.966T>A has been reported in the literature in at least one individual affected with Limb-Girdle Muscular Dystrophy (Senz_2005). These report(s) do not provide unequivocal conclusions about association of the variant with Limb-Girdle Muscular Dystrophy, Autosomal Recessive.
