This variant lies in the COL6A1 gene (transcript NM_001848.3) at coding-DNA position 2330 through coding-DNA position 2351, duplicating 22 bases; at the protein level this means shifts the reading frame starting at leucine residue 786, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This variant has not been reported in the literature in individuals affected with COL6A1-related conditions. ClinVar contains an entry for this variant (Variation ID: 1324136). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. This variant disrupts the C-terminus of the COL6A1 protein. Other variant(s) that disrupt this region (p.Leu941Alafs*34) have been observed in individuals with COL6A1-related conditions (PMID: 33250842).
