NM_004655.4(AXIN2):c.1713-2A>G was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.1713-2A>G intronic variant results from an A to G substitution two nucleotides upstream from coding exon 6 in the AXIN2 gene. This nucleotide position is conserved on limited sequence alignment. In silico splice site analysis predicts that this alteration will weaken the native splice acceptor site. RNA studies have demonstrated that this alteration results in an splice defect involving exons excluded from naturally occurring transcripts; the clinical impact of this abnormal splicing is unknown at this time (Ambry internal data). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
