NM_001292063.2(OTOG):c.2119dup (p.Glu707fs) was classified as Likely Pathogenic for Rare genetic deafness by Laboratory for Molecular Medicine, Mass General Brigham Personalized Medicine, citing ACMG Guidelines, 2015: The p.Glu719GlyfsX17 variant in OTOG has not been previously reported in individuals with nonsyndromic hearing loss and was absent from large population studies. This variant is predicted to cause a frameshift, which alters the protein’s amino acid sequence beginning at position 719 and leads to a premature termination codon 17 amino acids downstream. Loss of function of the OTOG gene is an established disease mechanism in autosomal recessive nonsyndromic hearing loss. In summary, although additional studies are required to fully establish its clinical significance, this variant meets criteria to be classified as likely pathogenic for autosomal recessive nonsyndromic hearing loss.
