NM_001025356.3(ANO6):c.1880+1G>A was classified as Uncertain significance for ANO6-related condition by PreventionGenetics, part of Exact Sciences, citing ACMG Guidelines, 2015. This variant lies in the ANO6 gene (transcript NM_001025356.3) at the canonical splice donor site of the intron immediately after coding-DNA position 1880, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: The ANO6 c.1880+1G>A variant is predicted to disrupt the GT donor site and interfere with normal splicing. To our knowledge, this variant has not been reported in the literature. This variant is reported in 0.013% of alleles in individuals of South Asian descent in gnomAD (http://gnomad.broadinstitute.org/variant/12-45797320-G-A). Upstream variants that disrupt consensus splice sites in ANO6 have been reported as pathogenic. Although we suspect that this variant may be pathogenic, at this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence.
