Pathogenic for Glycogen storage disease, type II — the classification assigned by Genomenon, Inc, Genomenon, Inc to NM_000152.5(GAA):c.309C>A (p.Cys103Ter), citing Genomenon Sequence Variant Interpretation Standards - Updated: GAA p.Cys103Ter (c.309C>A) is a nonsense variant that introduces a premature stop codon at amino acid position 103 and is predicted to result in a truncated or absent protein product. This variant has been observed in at least one proband with a GAA-related disorder (PMID:14695532). Functional studies have been reported (PMID:17805474). It is absent or not present at a significant frequency in gnomAD. In conclusion, we classify GAA p.Cys103Ter (c.309C>A) as a pathogenic variant.
