Likely pathogenic for Duchenne muscular dystrophy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004006.3(DMD):c.3430C>T (p.Gln1144Ter), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Gln1144*) in the DMD gene. RNA analysis indicates that this premature translational stop signal induces altered splicing and likely results in a shortened protein product. This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individuals with DMD-related conditions (PMID: 19937601, 31919629). ClinVar contains an entry for this variant (Variation ID: 1322364). Studies have shown that this premature translational stop signal results in skipping of exon 25, but is expected to preserve the integrity of the reading-frame (PMID: 31919629). In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic.
