Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_000051.4(ATM):c.4909G>A (p.Asp1637Asn), citing ACMG Guidelines, 2015. This variant lies in the ATM gene (transcript NM_000051.4) at coding-DNA position 4909, where G is replaced by A; at the protein level this means replaces aspartic acid at residue 1637 with asparagine — a missense variant. Submitter rationale: This variant causes a G to A nucleotide substitution at the last nucleotide of exon 32 of the ATM gene and replaces aspartic acid with asparagine at codon 1637 of the ATM protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with hereditary cancer in the literature. This variant has been identified in 1/251334 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
