Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001374736.1(DST):c.20205G>T (p.Glu6735Asp), citing Ambry Variant Classification Scheme 2023: The p.E4616D variant (also known as c.13848G>T), located in coding exon 76 of the DST gene, results from a G to T substitution at nucleotide position 13848. The glutamic acid at codon 4616 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
