This variant lies in the KMT2A gene (transcript NM_001197104.2) at coding-DNA position 134, where C is replaced by G; at the protein level this means replaces proline at residue 45 with arginine — a missense variant. Submitter rationale: Variant summary: KMT2A c.134C>G (p.Pro45Arg) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant was absent in 4836 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.134C>G in individuals affected with Wiedemann-Steiner Syndrome and no experimental evidence demonstrating its impact on protein function have been reported.
