Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001110556.2(FLNA):c.7379G>A (p.Gly2460Asp), citing Ambry Variant Classification Scheme 2023: The p.G2452D variant (also known as c.7355G>A), located in coding exon 44 of the FLNA gene, results from a G to A substitution at nucleotide position 7355. The glycine at codon 2452 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
