Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000075.4(CDK4):c.167A>C (p.Glu56Ala), citing Ambry Variant Classification Scheme 2023: The p.E56A variant (also known as c.167A>C), located in coding exon 1 of the CDK4 gene, results from an A to C substitution at nucleotide position 167. The glutamic acid at codon 56 is replaced by alanine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
