Pathogenic for Developmental and epileptic encephalopathy 94 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001271.4(CHD2):c.2759C>T (p.Thr920Ile), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces threonine with isoleucine at codon 920 of the CHD2 protein (p.Thr920Ile). The threonine residue is highly conserved and there is a moderate physicochemical difference between threonine and isoleucine. This variant is not present in population databases (ExAC no frequency). This missense change has been observed in individual(s) with clinical features of childhood-onset epileptic encephalopathy (Invitae). In at least one individual the variant was observed to be de novo. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt CHD2 protein function. For these reasons, this variant has been classified as Pathogenic.
