Uncertain Significance for Ehlers-Danlos syndrome, type 4 — the classification assigned by All of Us Research Program, National Institutes of Health to NM_000090.4(COL3A1):c.23G>A (p.Gly8Glu), citing ACMG Guidelines, 2015. This variant lies in the COL3A1 gene (transcript NM_000090.4) at coding-DNA position 23, where G is replaced by A; at the protein level this means replaces glycine at residue 8 with glutamic acid — a missense variant. Submitter rationale: This variant is absent from or rare in large population databases, including the Genome Aggregation Database (http://gnomad.broadinstitute.org/). To date, this variant has not been reported in the literature in individuals with vEDS. This variant is not expected to disrupt protein function.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
