NM_001378454.1(ALMS1):c.5860G>A (p.Glu1954Lys) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E1955K variant (also known as c.5863G>A), located in coding exon 8 of the ALMS1 gene, results from a G to A substitution at nucleotide position 5863. The glutamic acid at codon 1955 is replaced by lysine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
