NM_004722.4(AP4M1):c.1360T>C (p.Ter454Arg) was classified as Uncertain significance for Hereditary spastic paraplegia 50 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change disrupts the translational stop signal of the AP4M1 mRNA. It is expected to extend the length of the AP4M1 protein by 84 additional amino acid residues. This variant is not present in population databases (gnomAD no frequency). This protein extension has been observed in individual(s) with clinical features of hereditary spastic paraplegia (PMID: 41018996; internal data). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 1309955). Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
