NM_000548.5(TSC2):c.381C>G (p.Ile127Met) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.I127M variant (also known as c.381C>G), located in coding exon 4 of the TSC2 gene, results from a C to G substitution at nucleotide position 381. The isoleucine at codon 127 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
