Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_003036.4(SKI):c.1870G>C (p.Glu624Gln), citing Ambry Variant Classification Scheme 2023: The p.E624Q variant (also known as c.1870G>C), located in coding exon 6 of the SKI gene, results from a G to C substitution at nucleotide position 1870. The glutamic acid at codon 624 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
