Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000238.4(KCNH2):c.567_575dup (p.Ala193_Val194insProGlyAla), citing Ambry Variant Classification Scheme 2023. This variant lies in the KCNH2 gene (transcript NM_000238.4) at coding-DNA position 567 through coding-DNA position 575, duplicating 9 bases. Submitter rationale: The c.567_575dupCGCCCCGGG variant (also known as p.A193_V194insAPG), located in coding exon 4 of the KCNH2 gene, results from an in-frame duplication of CGCCCCGGG at nucleotide positions 567 to 575. This results in the duplication of 3 extra residues (APG) between codons 193 and 194. This region is not well conserved in available vertebrate species. In addition, the in silico prediction for this variant is inconclusive (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
