This variant lies in the IFIH1 gene (transcript NM_022168.4) at coding-DNA position 2398, where G is replaced by T; at the protein level this means converts the codon for glutamic acid at residue 800 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This sequence change creates a premature translational stop signal (p.Glu800*) in the IFIH1 gene. It is expected to result in an absent or disrupted protein product. However, the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in IFIH1 cause disease. This variant is present in population databases (no rsID available, gnomAD 0.006%). This variant has not been reported in the literature in individuals affected with IFIH1-related conditions. This premature translational stop signal has been observed in at least one individual who was not affected with IFIH1-related conditions (internal data). ClinVar contains an entry for this variant (Variation ID: 1306303). In summary, the available evidence is currently insufficient to determine the role of this variant in disease.
