Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NC_000015.10:g.84817253C>G, citing Ambry Variant Classification Scheme 2023: The p.S136C variant (also known as c.407C>G), located in coding exon 1 of the ALPK3 gene, results from a C to G substitution at nucleotide position 407. The serine at codon 136 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
