Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004655.4(AXIN2):c.1031A>C (p.Asn344Thr), citing Ambry Variant Classification Scheme 2023: The p.N344T variant (also known as c.1031A>C), located in coding exon 3 of the AXIN2 gene, results from an A to C substitution at nucleotide position 1031. The asparagine at codon 344 is replaced by threonine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
