Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000090.4(COL3A1):c.1102A>G (p.Arg368Gly), citing Ambry Variant Classification Scheme 2023: The p.R368G variant (also known as c.1102A>G), located in coding exon 16 of the COL3A1 gene, results from an A to G substitution at nucleotide position 1102. The arginine at codon 368 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
