This variant lies in the RYR1 gene (transcript NM_000540.3) at coding-DNA position 9848, where G is replaced by A; at the protein level this means replaces arginine at residue 3283 with glutamine — a missense variant. Submitter rationale: This missense variant replaces arginine with glutamine at codon 3283 of the RYR1 protein. Computational prediction is inconclusive regarding the impact of this variant on protein structure and function (internally defined REVEL score threshold 0.5 < inconclusive < 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in one individual who experienced a fatal malignant hyperthermia event and was also found to carry a variant of uncertain significance in CACNA1S (PMID: 23558838, 25268394). This variant has been identified in 5/249582 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
