Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_198586.3(NHLRC1):c.773A>G (p.Gln258Arg), citing Ambry Variant Classification Scheme 2023: The p.Q258R variant (also known as c.773A>G), located in coding exon 1 of the NHLRC1 gene, results from an A to G substitution at nucleotide position 773. The glutamine at codon 258 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
