Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000539.3(RHO):c.568G>A (p.Asp190Asn), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces aspartic acid, which is acidic and polar, with asparagine, which is neutral and polar, at codon 190 of the RHO protein (p.Asp190Asn). This variant is present in population databases (rs104893779, gnomAD 0.006%). This missense change has been observed in individual(s) with autosomal dominant retinitis pigmentosa (PMID: 1765377, 19085385). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 13023). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. However, the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on RHO protein function.
