Pathogenic for Trichohepatoenteric syndrome 2 — the classification assigned by Dubai Health Genomic Medicine Center, Dubai Health to NM_006929.5(SKIC2):c.2341-1G>T, citing ACMG Guidelines, 2015: The c.2341-1G>T variant in SKIV2L has not been previously reported in individuals with disease and was absent from large population studies such as the Genome Aggregation Database (gnomAD) and the Greater Middle East (GME) variome database. This variant occurs in the invariant region (+/- 1/2) of the splice consensus sequence and is predicted to cause altered splicing leading to an abnormal or absent protein. Other disease causing variants have been reported in the same exon where the c.2341-1G>T is found (Human Gene Mutation Database). In summary this variant meets our criteria to be classified as pathogenic.
