Likely pathogenic for Charcot-Marie-Tooth disease type 4B3 — the classification assigned by Dubai Health Genomic Medicine Center, Dubai Health to NM_002972.4(SBF1):c.5463C>G (p.Tyr1821Ter), citing ACMG Guidelines, 2015: The Tyr1821* variant in SBF1 has not been previously reported in patients and was absent from large population studies such as the Genome Aggregation Database (gnomAD) and the Greater Middle East (GME) Variome database. This nonsense variant leads to a premature termination codon at position 1821 which is then predicted to lead to a truncated or absent protein. In summary this variant meets our criteria to be classified as likely pathogenic.
