This variant lies in the ATP8B1 gene (transcript NM_001374385.1) at coding-DNA position 2006 through coding-DNA position 2007, replacing the reference sequence with CA; at the protein level this means replaces tryptophan at residue 669 with serine — a missense variant. Submitter rationale: Variant summary: ATP8B1 c.2006_2007delinsCA (p.Trp669Ser) results in a non-conservative amino acid change located in the P-type ATPase, haloacid dehalogenase domain (IPR044492) of the encoded protein sequence. Four of four in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251242 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.2006_2007delinsCA in individuals affected with Familial Intrahepatic Cholestasis and no experimental evidence demonstrating its impact on protein function have been reported.
