NM_198253.3(TERT):c.-124C>T was classified as Uncertain significance by Genetic Services Laboratory, University of Chicago, citing ACMG Guidelines, 2015: DNA sequence analysis of the TERT gene demonstrated a sequence change in the promoter region, c.-124C>T. This change does not appear to have been previously described in individuals with TERT-related disorders; however it has been reported at a relatively high frequency in tumor tissue samples including thyroid cancer, urological malignancies, etc. (PMID: 31669704, 31408918). This alteration was reported to lead to a significant increase in TERT promoter activity when assayed using luciferase reporter system (PMID: 31408918). This sequence change has been described in the gnomAD database with an overall frequency of 0.0095% (dbSNP rs1242535815). The functional significance of this sequence change in a germline sample is unclear and its contribution to this individual's disease phenotype cannot definitively be determined
