Uncertain significance for Retinitis pigmentosa — the classification assigned by Broad Center for Mendelian Genomics, Broad Institute of MIT and Harvard to NM_003322.6(TULP1):c.1597T>C (p.Ser533Pro), citing ACMG Guidelines, 2015. This variant lies in the TULP1 gene (transcript NM_003322.6) at coding-DNA position 1597, where T is replaced by C; at the protein level this means replaces serine at residue 533 with proline — a missense variant. Submitter rationale: The p.Ser533Pro variant in TULP1 was identified in an individual with Retinitis pigmentosa, via a collaborative study between the Broad Institute's Center for Mendelian Genomics and the Pierce lab (https://oculargenomics.meei.harvard.edu/labs/pierce-lab/lab-members/). Through a review of available evidence we were able to apply the following criteria: PM2, PP3. Based on this evidence we have classified this variant as a Variant of Uncertain Significance. If you have any questions about the classification please reach out to the Pierce Lab.
