NM_000540.3(RYR1):c.1209C>G (p.Ile403Met) was classified as Uncertain significance by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015: Variant summary: RYR1 c.1209C>G (p.Ile403Met) results in a conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 251444 control chromosomes. c.1209C>G has been reported in the literature in heterozygous individuals affected with Central Core Disease (Quane_1993, Fusto_2022) and an individual with a family history of Malignant Hyperthermia Susceptibility (Ibarra Moreno_2020). These data do not allow any conclusion about variant significance. At least one publication reports experimental evidence evaluating an impact on protein function. The most pronounced variant effect results in increased sensitivity to RYR1 agonists in transfected HEK293 cells (Tong_1997). The following publications have been ascertained in the context of this evaluation (PMID: 11524458, 35428369, 32861507, 8220423, 9334205, 9873004). ClinVar contains an entry for this variant (Variation ID: 12968). Based on the evidence outlined above, the variant was classified as VUS-possibly pathogenic.
