NM_000094.4(COL7A1):c.6103del (p.Glu2035fs) was classified as Pathogenic for Recessive dystrophic epidermolysis bullosa by Department of Pediatrics, Division of Medical Genetics, Faculty of Medicine Ramathibodi Hospital, Mahidol University. This variant lies in the COL7A1 gene (transcript NM_000094.4) at coding-DNA position 6103, deleting one base; at the protein level this means shifts the reading frame starting at glutamic acid residue 2035, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: Heterozygous of c.6103delG variants in COL7A1 gene was identified in the patient with Epidermolysis bullosa Epidermolysis bullosa dystrophica inversa, and was absent from large population studies (genomAD database). The c.6103delG variant meets ACMG criteria to be classified as pathogenic based upon null variant, absent from controls, multiple lines of computational evidence support a deleterious effect on the gene, detected in trans with a pathogenic variant, and patient’s phenotype or family history is highly specific for the disease.
