Benign — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000397.4(CYBB):c.484-60TG[21], citing LabCorp Variant Classification Summary - May 2015: Variant summary: CYBB c.484-17_484-14delGTGT alters a nucleotide located at a position not widely known to affect splicing. Consensus agreement among computation tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 0.042 in 15161 control chromosomes, predominantly at a frequency of 0.097 within the African or African-American subpopulation in the gnomAD database, including 42 homozygotes. The observed variant frequency within African or African-American control individuals in the gnomAD database exceeds the estimated maximal expected allele frequency for disease-causing variants in CYBB. To our knowledge, no occurrence of c.484-17_484-14delGTGT in individuals affected with CYBB-related conditions and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 1289783). Based on the evidence outlined above, the variant was classified as benign.
