This variant lies in the CYP27A1 gene (transcript NM_000784.4) at coding-DNA position 203, where T is replaced by G; at the protein level this means replaces phenylalanine at residue 68 with cysteine — a missense variant. Submitter rationale: Variant summary: CYP27A1 c.203T>G (p.Phe68Cys) results in a non-conservative amino acid change in the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 251328 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, c.203T>G has not been reported in the literature in individuals affected with Cerebrotendinous Xanthomatosis and no experimental evidence demonstrating an impact on protein function has been reported.
