NM_016203.4(PRKAG2):c.253C>T (p.Pro85Ser) was classified as Uncertain Significance for Hypertrophic cardiomyopathy by All of Us Research Program, National Institutes of Health, citing ACMG Guidelines, 2015: This missense variant replaces proline with serine at codon 85 of the PRKAG2 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in two individuals affected with hypertrophic cardiomyopathy; one of these individuals also carried a pathogenic truncating variant in the MYBPC3 gene (PMID: 23785128, 30847666). This variant has also been reported in an individual affected with dilated cardiomyopathy and in an individual affected with unknown arrhythmia (PMID: 30847666). This variant has been identified in 2/245228 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
