This variant lies in the MYBPC3 gene (transcript NM_000256.3) at coding-DNA position 2864, where C is replaced by A; at the protein level this means replaces proline at residue 955 with histidine — a missense variant. Submitter rationale: The p.P955H variant (also known as c.2864C>A), located in coding exon 27 of the MYBPC3 gene, results from a C to A substitution at nucleotide position 2864. The proline at codon 955 is replaced by histidine, an amino acid with similar properties. This variant was detected in a cardiomyopathy/arrhythmia genetic testing cohort; however, clinical details were limited, and additional cardiac variants were detected in some cases (van Lint FHM et al. Neth Heart J, 2019 Jun;27:304-309). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
