Uncertain significance for Leber congenital amaurosis 13 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_152443.3(RDH12):c.530C>T (p.Ala177Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces alanine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 177 of the RDH12 protein (p.Ala177Val). This variant is present in population databases (rs373032226, gnomAD 0.1%). This missense change has been observed in individual(s) with cone-rod dystrophies (PMID: 17512964). ClinVar contains an entry for this variant (Variation ID: 1284500). Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The valine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. Experimental studies have shown that this missense change does not substantially affect RDH12 function (PMID: 17512964). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
