This variant lies in the ABRAXAS1 gene (transcript NM_139076.3) at coding-DNA position 364, where C is replaced by T; at the protein level this means converts the codon for glutamine at residue 122 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: Variant summary: FAM175A c.364C>T (p.Gln122X) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, which are commonly known mechanisms for disease. The variant allele was found at a frequency of 3.2e-05 in 251138 control chromosomes, predominantly at a frequency of 6.2e-05 within the Non-Finnish European subpopulation in the gnomAD database. c.364C>T has been reported in the literature in individuals affected with advanced head and neck cancer (Bertelsen_2019). To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
