NM_005732.4(RAD50):c.3467G>A (p.Arg1156His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R1156H variant (also known as c.3467G>A), located in coding exon 22 of the RAD50 gene, results from a G to A substitution at nucleotide position 3467. The arginine at codon 1156 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
