Pathogenic — the classification assigned by GeneDx to NM_004329.3(BMPR1A):c.369del (p.Glu123fs), citing GeneDx Variant Classification (06012015): This pathogenic variant is denoted BMPR1A c.369delA at the cDNA level and p.Glu123AspfsX21 (E123DfsX21) at the protein level. The normal sequence, with the base that is deleted in brackets, is TAGA(delA)TGTT. The deletion causes a frameshift, which changes a Glutamic Acid to an Aspartic Acid at codon 123, and creates a premature stop codon at position 21 of the new reading frame. This variant is predicted to cause loss of normal protein function through either protein truncation or nonsense-mediated mRNA decay. Although this variant has not been previously reported to our knowledge, it is considered pathogenic.
