NM_000051.4(ATM):c.2450A>T (p.Asp817Val) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015): This variant is denoted ATM c.2450A>T at the cDNA level, p.Asp817Val (D817V) at the protein level, and results in the change of an Aspartic Acid to a Valine (GAT>GTT). This variant has not, to our knowledge, been published in the literature as pathogenic or benign. ATM Asp817Val was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. This variant is a non-conservative substitution in which a negative polar amino acid is replaced with a neutral non-polar one, altering a position that is well conserved throughout evolution and is not located in a known functional domain. Multiple in silico algorithms predict that this variant may be damaging to protein structure and function. Based on the currently available information, we consider ATM Asp817Val to be a variant of uncertain significance.
