Pathogenic for Hereditary breast and ovarian cancer syndrome — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000059.4(BRCA2):c.5863del (p.Ser1955fs), citing LabCorp Variant Classification Summary - May 2015: Variant summary: BRCA2 c.5863delT (p.Ser1955GlnfsX8) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, which are commonly known mechanisms for disease. The variant was absent in 250960 control chromosomes (gnomAD). c.5863delT has been reported in the literature in a family at risk for breast and/or ovarian cancer (Rebbeck_2018). To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Five other ClinVar submitters (evaluation after 2014) including an expert panel (ENIGMA) cite the variant as pathogenic. Based on the evidence outlined above, the variant was classified as pathogenic.
